Overview
Goldenrod’s lead candidate, 11h, is a next‑generation, orally bioavailable, brain‑penetrant small‑molecule PDE4 inhibitor in‑licensed from the University of Nebraska Medical Center. 11h was designed to overcome limitations of earlier PDE4 inhibitors by improving CNS exposure and minimizing emetic responses in preclinical studies. The company’s initial clinical strategy is a Phase I study in Friedreich’s Ataxia to evaluate safety, tolerability, pharmacokinetics and pharmacodynamic biomarkers, with clinical development expected to begin in 2027. Goldenrod says positive Phase I biomarker and PK data would inform dose selection and enable development across additional neurodegenerative and neuroinflammatory indications, including Alzheimer’s disease, MS, ALS, substance use disorders, Batten disease, pain and traumatic brain injury. Development of 11h has been supported by grant funding from the NIH and the Department of Defense as well as funding from Fannin.
