Overview

Tenaya Therapeutics develops potentially curative therapies that target underlying causes of heart disease through three platforms: Gene Therapy, Cellular Regeneration and Precision Medicine. Its lead program is an AAV gene therapy targeting MYBPC3-linked hypertrophic cardiomyopathy, currently in IND-enabling studies. The company is also developing a DWORF AAV gene therapy, an in vivo cellular reprogramming approach using a single AAV vector to convert fibroblasts into cardiomyocytes, and a small-molecule program with completed medicinal chemistry and filed IP. Tenaya has built in-house capabilities for AAV capsid engineering, proprietary regulatory elements, disease modeling using iPSC-derived cardiomyocytes, and has filed related IP. It is internalizing AAV manufacturing with a 200L pilot plant and plans a 1,000L cGMP facility in the San Francisco Bay Area to support first-in-human studies. Tenaya Therapeutics is advancing multiple therapeutic candidates across three core product platforms: Cellular Regeneration, Gene Therapy, and Precision Medicine. The Cellular Regeneration platform uses novel AAV vectors to deliver proprietary transcription factors for in vivo reprogramming of cardiac fibroblasts into cardiomyocytes, initially targeting acute and chronic injury after myocardial infarction. The Gene Therapy platform employs AAV vectors for targeted delivery and expression of therapeutic payloads to specific heart cells, focusing on genetically defined cardiomyopathies. The Precision Medicine platform uses isogenic iPSC-derived cardiomyocytes as human disease models to identify and validate heart-failure targets and to screen therapeutic compounds, with an initial focus on small molecules for genetically defined dilated cardiomyopathies. The company intends to use the Series B proceeds to advance and expand its pipeline and to strengthen the underlying science, capabilities, and intellectual property for its multi-modality platforms. Tenaya was founded by scientists from the Gladstone Institute’s Cardiovascular Division and UT Southwestern Medical Center and is led by CEO Faraz Ali; it has raised $142M since its founding.
