Treadwell Therapeutics was launched by TIO Bioventures to develop first-in-class and best-in-class oral small-molecule therapies that exploit cancer cell vulnerabilities and immune-oncology targets. The company’s internally developed pipeline includes a first-in-class PLK4 inhibitor (CFI-400945) and a potentially best-in-class TTK inhibitor (CFI-402257) that have advanced from discovery to Phase 2 and shown encouraging safety and efficacy in investigator-sponsored trials. A third molecule, CFI-402411—an oral immunomodulatory kinase inhibitor targeting HPK1—was planned to enter the clinic in early 2020 with preclinical data supporting monotherapy and combination use with checkpoint inhibitors. Treadwell’s product candidates are wholly owned and were developed within TIO Discovery, the R&D engine of TIO Bioventures. The company is supported operationally and strategically by TIO Discovery’s drug discovery and early clinical development capabilities and a leadership team with extensive oncology development experience.
