Sigilon develops Shielded Living Therapeutics: engineered human cells encapsulated in its Afibromer biomaterials matrix to produce therapeutic proteins while avoiding immune rejection and fibrosis. The company’s platform targets chronic diseases including hemophilia, diabetes, lysosomal disorders and other endocrine and immune conditions. Its lead investigational therapy, SIG‑001 for hemophilia A, has received Orphan Drug Designation from the FDA. Sigilon plans to initiate a first‑in‑human clinical trial of SIG‑001 in the first half of 2020 and to continue advancing and expanding programs across rare blood disorders, lysosomal diseases and endocrine and immune disorders. Financially, the company completed an $80.3 million Series B that brings total funding to more than $195 million. Sigilon was founded by Flagship Pioneering in conjunction with Daniel Anderson, Ph.D., and Robert Langer, Sc.D., and is based in Cambridge, Mass. Sigilon develops immune‑privileged living therapeutic implants based on its Afibromer™ technology platform, focused on encapsulated cell therapies. The company announced a collaboration with Eli Lilly to develop therapies for type 1 diabetes, under which Lilly receives an exclusive worldwide license to Sigilon’s Afibromer technology for islet cell encapsulation. Sigilon will lead and fund development activities through submission of an IND, after which Lilly will assume responsibility for clinical development and commercialization. Financial terms include a $63M upfront payment, an undisclosed equity investment from Lilly, and eligibility for up to $410M in development and commercialization milestones plus single- to double-digit tiered royalties on future product sales. The company was unveiled by Flagship Pioneering in 2017 with $23.5M in initial capital and is led by CEO Paul Wotton, Ph.D.
