Third Harmonic Bio is a clinical-stage biotech advancing THB001, a first-in-class, highly selective oral small-molecule KIT inhibitor designed to deplete mast cells and blunt mast cell–driven inflammation. In preclinical studies THB001 showed promising drug-like properties, excellent kinome selectivity, and potent mast cell depletion across relevant tissues. Early clinical data demonstrated robust pharmacodynamic effects measured by serum tryptase. The company plans to initiate a Phase 1b proof-of-concept study in cold-inducible urticaria in the second half of 2022 and intends to pursue additional dermatologic, respiratory, and gastrointestinal mast cell–mediated indications. The recently closed financing is expected to fund THB001 through the next phase of human studies for chronic urticaria.
