OrsoBio is advancing a portfolio of mitochondrial protonophores designed to increase energy expenditure and complement GLP-1 RA–based weight-loss therapies. Its lead oral, liver-targeted candidate TLC-6740 is in a 12-week Phase 1b trial in patients with obesity, and a prior Phase 1 in healthy subjects showed safety, dose-dependent increases in energy expenditure, and improvements in lipids and insulin-resistance markers. IND-enabling studies for two additional protonophores (TLC-1180 and TLC-1235) with distinct distribution and delivery attributes are expected to complete in 2025. TLC-1180 is described as a potent, long-acting protonophore suitable for weekly oral or subcutaneous dosing; TLC-1235 is a controlled-release mitochondrial protonophore. OrsoBio also has TLC-3595 (an ACC2 inhibitor) and TLC-2716 (an LXR inverse agonist) in Phase 2a studies for type 2 diabetes, severe hypertriglyceridemia, and MASH. The company intends to use newly raised capital to accelerate clinical development across these programs.
