Leal Therapeutics focuses on developing neuro-metabolic treatments for high-need central nervous system disorders and was launched in 2021. Its lead program, LTX-001, is a brain-penetrant oral GLS1 inhibitor that completed single and multiple ascending dose Phase 1 trials and has initiated a Phase 1b/2a trial in adults with schizophrenia with initial data expected by year-end. A second program, LTX-002, is an intrathecally delivered antisense oligonucleotide targeting SPTLC1 and is being evaluated in the NeurALS Phase 1/2 trial in adults with ALS. The company also is developing a next-generation CNS delivery platform using antibody-like shuttle technology to enable transport of peripherally administered therapeutics to the brain. Leal states it has capabilities to develop and produce both small molecule and nucleic acid CNS therapeutics. The recent Series A second close of $30 million, which added Eli Lilly to its investor syndicate, is intended to support clinical milestones for LTX-001 and additional dosing cohorts for LTX-002.

Ten63 Therapeutics is developing BEYOND, a Large Quantum Chemistry Model (LQCM) that combines physics-based simulation with artificial intelligence to explore trillions of potential small-molecule drug candidates per protein target with near-experimental accuracy. The platform is designed to crack the 80 % of human proteins long deemed “undruggable,” opening new therapeutic avenues in oncology, infectious disease, and other areas. With financial backing, the company is prioritizing programs such as affordable treatments for human papillomavirus (HPV) in partnership with the Gates Foundation. Ten63 positions its technology as moving AI drug discovery from decision-support toward true hypothesis generation and “superintelligence.” Located in North Carolina’s Research Triangle Park, the company benefits from local computational biology and biopharma infrastructure. To date, Ten63 has raised more than $45 million, including a newly announced strategic investment. The fresh capital will fund further scaling of the BEYOND platform and advancement of internal drug programs.

Founded in 2023, Dualitas Therapeutics is building a pipeline of bispecific antibodies that simultaneously engage two immune-cell targets to achieve greater efficacy than single-target monoclonal antibodies. Its internally developed DualScreen platform scans the entire cell surface (“surfaceome”) to identify hundreds of thousands of target pairings and select the most promising therapeutic combinations using a proximity-biology approach. The two lead preclinical candidates are DTX-103, aimed first at asthma and other allergic diseases, and DTX-102, initially focused on rheumatoid arthritis; specific targets for both remain undisclosed. One of these programs is expected to be advanced in-house, while the other may be partnered with a larger pharmaceutical company. Management plans to move the internal lead into clinical trials within 18–24 months and will evaluate clinical differentiation versus current standards of care to guide trial design. The company’s scientific leadership includes co-founders Forbes Huang (COO/CBO) and Greg Lazar (CSO), with Rich Murray serving as interim CEO.

Stylus Medicine develops transformative in vivo genetic medicines by combining sequence-specific, therapeutic-grade recombinases with cell-targeted lipid nanoparticle (LNP) delivery. Its platform is designed to encode polyfunctional multi-kilobase payloads with high efficiency, specificity, and fidelity, enabling durable and scalable in vivo cell engineering. The approach aims to overcome limitations of ex vivo and viral manufacturing and to simplify patient treatment workflows. Stylus intends to start by enabling in vivo CAR-T therapies and positions its technology for applications across oncology, autoimmune, and genetic diseases. The company emerged from stealth with $85 million in financing and will use the proceeds to advance its platform and pipeline of in vivo therapeutic programs. Stylus is led by industry veterans including Chairman and CEO Emile Nuwaysir and CSO Jason Fontenot and is based in Cambridge, Massachusetts; it was created in 2022 by Raven, RA Capital’s incubator, and Khosla Ventures.
